State Route 388 (SR 388) is a 4.5-mile-long (7.2 km) highway that runs south-to-north, completely within Columbia County, in the American state of Georgia.

SR 388 runs from SR 223 in Grovetown, in the south, to SR 232 in the community of Lewiston, in the north, along Wrightsboro Road, Horizon South Parkway, and Lewistown Road. The route has an interchange with I-20 (unsigned SR 402) near its midpoint. SR 388 serves to connect SR 223 and SR 232, which run parallel to each other on opposite sides of I-20 in southern Columbia County.
