State Route 43 (SR 43) is a state highway that runs south-to-north through portions of McDuffie and Lincoln counties in east-central Georgia. The route runs from its southern terminus at US 78, SR 10 and SR 17 north of Thomson to its northern terminus at US 378 and the South Carolina state line northeast of Lincolnton.

From its southern terminus at US 78/SR 10/SR 17 north of Thomson in McDuffie County, SR 43 travels north. It passes over a branch of Strom Thurmond Lake, and enters Lincoln County. The route intersects SR 220 in the southern portion of the county, and then continues north to Lincolnton, the county seat. In Lincolnton, SR 43 briefly runs concurrent with SR 47. The two routes travel a short distance west to meet US 378. At the intersection, State Routes 43 and 47 diverge; SR 43 becomes concurrent with US 378 northeast of the city, and SR 47 becomes concurrent with US 378 west of the city. US 378/SR 43 remain concurrent to the South Carolina state line, where SR 43 meets its northern terminus and US 378 continues to McCormick, South Carolina.

SR 43 is a rural route, serving mostly to connect the cities of Lincolnton and Thomson. The route sees an Average Annual Daily Traffic (AADT) of less than 5,000 vehicles over its entire length.
